چکیده

An in vivo thymus reconstitution assay based on intrathymic injection of hematopoietic progenitors into irradiated chicks was used to determine the number of T-cell progenitors in peripheral blood, paraaortic foci, bone marrow (BM), and spleen during ontogeny. This study allowed us to analyze the regulation of thymus colonization occurring in three waves during embryogenesis. It confirmed that progenitors of the first wave of thymus colonization originate from the paraaortic foci, whereas progenitors of the second and the third waves originate from the BM. The analysis of the number of T-cell progenitors indicates that each wave of thymus colonization is correlated with a peak number of T-cell progenitors in peripheral blood, whereas they are almost absent during the periods defined as refractory for colonization. Moreover, injection of T-cell progenitors into the blood circulation showed that they homed into the thymus without delay during the refractory periods. Thus, thymus colonization kinetics depend mainly on the blood delivery of T-cell progenitors during embryogenesis. r 1999 by The American Society of Hematology.

Early ontogeny of thymocytes in pigs: sequential colonization of the thymus by T cell progenitors.

Successive colonization of the thymus by waves of thymocyte progenitors has been described in chicken-quail chimeras and suggested from studies in mice. In swine, we show that the first CD3epsilon-bearing thymocytes appear on day 40 of gestation (DG40). These early thymocytes were CD3epsilonhigh and belonged to the gammadelta T cell lineage. Mature CD3epsilonhigh alphabeta thymocytes were obser...

Effects of Recombinant Human Erythropoietin (rHuEpo) on the Blood Cells and Hematopoiesis in Spleen of Rats

Recombinant human erythropoietin is a glycoprotein hormone that stimulates erythropoiesis. In clonogenic assays of hematopoietic progenitors, high concen-trations of erythropoietin (Epo) increase CFU-E and diminish the number of granulocytes formed per culture plate. Fetal progenitors are more sensitive to these effects of Epo than progenitors from adults. We administered doses 50, 100, 200 IU/...

Multicongenic fate mapping quantification of dynamics of thymus colonization

Postnatal T cell development depends on continuous colonization of the thymus by BM-derived T lineage progenitors. Both quantitative parameters and the mechanisms of thymus seeding remain poorly understood. Here, we determined the number of dedicated thymus-seeding progenitor niches (TSPNs) capable of supporting productive T cell development, turnover rates of niche occupancy, and feedback mech...

Ontogeny of the Immune System: γ/δ and α/β T Cells Migrate from Thymus to the Periphery in Alternating Waves

The embryonic thymus is colonized by the influx of hemopoietic progenitors in waves. To characterize the T cell progeny of the initial colonization waves, we used intravenous adoptive transfer of bone marrow progenitors into congenic embryos. The experiments were performed in birds because intravenous cell infusions can be performed more efficiently in avian than in mammalian embryos. Progenito...
